Tissue Culture Acclimation Guide
Materials Needed
• Tissue culture plantlets
• Clean water (preferably distilled or filtered)
• 70% isopropyl alcohol
• Rooting hormone (provided in starter set)
• Brownie Plugs (provided in starter set)
• Domed seed tray (provided in starter set)
Step 1: Pre-Soak the Brownie Plugs
1. Fill a large bowl with brownie plugs and clean water.
Step 2: Sanitize Tools and Workspace
1. Wipe your tools and working surface with 70% isopropyl alcohol.
2. Allow tools to air dry before use.
This helps prevent contamination while the plants are still vulnerable.
Step 3: Remove Plant from Culture Medium
1. Gently remove the plantlet from the tissue culture bag.
2. Handle the plant carefully by the leaves or base, avoiding damage to the roots.
Step 4: First Rinse
1. Rinse the plant gently under lukewarm running water.
2. Carefully remove all remaining agar or gel from the roots.
Agar left on roots can encourage mold or bacteria.
Step 5: Sanitation Rinse
1. Dunk the plants in 70% isopropyl alcohol for 10 seconds to reduce microbial contamination.
2. Do not soak the plant—just a brief rinse is sufficient.
Step 6: Final Water Rinse
1. Rinse the plant again thoroughly with clean water.
2. This removes residual alcohol and reduces the risk of root damage.
Step 7: Apply Rooting Hormone
1. Lightly dip the roots in the rooting hormone powder.
2. Tap off excess powder.
This can encourage faster root development in the new substrate.
Step 8: Planting
1. Squeeze excess water from a brownie plug.
2. Rip the plug down the middle vertically.
3. Gently sandwich the plant in the plug and place in the tray.
Avoid burying leaves or the crown of the plant.
Step 9: Create the Humidity Environment
1. Place the domed lid on securely.
2. Make sure the vents are fully closed.
Tissue culture plants lack a protective cuticle and need humidity while adjusting.
Step 10: Light and Temperature
• Place the domed tray in bright, indirect light.
• Avoid direct sun, which can overheat the plants.
• Maintain temperatures around 70–80°F (21–27°C).
Step 11: Gradual Acclimation
After about 14 days:
1. Begin opening the vents slightly each day.
2. Increase ventilation over 1–2 weeks.
3. Eventually remove the domed lid completely once the plant shows stable growth and you see new roots growing in the tray cells.
Step 12: Monitoring
Watch for:
• Wilting (needs water or more humidity)
• Mold growth (too much water or humidity)
• Yellowing leaves (older leaves may fall off)
If mold appears, increase airflow and remove affected material.
Step 13: Potting Up
After a week or so of your plants being fully acclimated to ambient air (no domed lid) it is safe to pot your new plants into a 4” pot. Use a well draining mix. We like to mix coir, perlite, and orchid bark. Our fertilizer of choice is Superthrive Foliage Pro.
Tip: Newly acclimated plants grow slowly at first. Once established, they can be transplanted into a more standard growing mix if needed.
